Do subs need to break in?

So my other question is...How much of a difference will there be going from a sealed 1.3cu ft box to a ported box tuned at 33hz?

It should make a significant difference. Like i said, it will let the woofer reach its full potential with less power. If the 500d really puts out 600w (I've had one and I will agree with what Memphis told you;) ), then you should be close to the RL-p's potential. How much difference there will be between the type s and the RL-p, I can't tell you. I haven't heard either. But, generally speaking, switching to the bigger sub and a larger, ported enclosure should yield a nice increase in output.

for me going from a .75cu.ft to a 2cu.ft 32hz was like 10db..

Ported = better.
I will agree that ported is a more efficient enclosure design than sealed as it takes less power to get loud. FACT: Louder does NOT mean BETTER. In my 15 years of car audio I've always preferred the sound of a sealed enclosure over a ported. It is a matter of taste so if one person likes his loud at all costs then ported would definitely be the way to go.

 
You bought the wrong sub to get loud. If thats all you wanted you could have spent your money better. Although with a good amount of power the RLP is nothing to laugh at.

 
I'd turn your gains up. Remeber, your putting out max power at 80% on your volume with a 0db test tone. You'll never see full power out of your amp on music, ever, not with that setting. Sure, you'll never clip, but it's like having a ferrari and then putting a govenor on it and wondering why it's not going fast. If you don't want to set by ear use a -6db test tone. You'll see quite a bit more power out of your amp, you will only clip slightly at high volume trasients meaning at high volume levels with bass heavy music, but unless you plan on listening to really heavy stuff all the time (like a 0db test tone) you wont' clip. It's much more realistic for the intended purpose of the sub (playing music).

 
To answer the very original question though, YES, subs do require break-in before performing optimally. But it wont be a night and day difference, so don't count on it to sound any louder with time. Better perhaps, but not louder.

All speakers have break-in, because there are moving parts.
