Setting gains with DMM, probing speaker power cable?

Hi, I would like to know this, I want to set the gains of my amp using a DMM, and I want to know if instead of probing the speaker outputs, if i can probe the speaker wire connected to my box, it should be the same i think. I want to do this because it is a PITA to take out the seat to take of the speaker wire.

yup, thats fine. the speaker wire and outputs at the amp are basically all one "node" so you can probe either
