Questions about where a port is located?

hey guys i know this may sound very stupid but i was jsut curious if where a port is placed on the box if it changes how the subs will sound. for instance in my truck its an extended cab and i limited on space. now would it make a difference if they made the box for me with the port on the side faces the driver side wall or if they made the box with the port coming up top towards the roof of my truck. would it affect that subs at all?
