No you don't put sound deadening inside the enclosure.

Sound deadening is for controlling panel resonance not to add rigidity. It's designed for sheet metal and other thin substrates not wood.

Ideally you want the enclosure to be as hard as possible. You can reinforce the interior with wood hardener or fiberglass resin but it's really unnecessary unless you are in a competition and chasing every tenth of spl possible.

I would face the port and sub back. Even if you got a soft top, the rear gate will serve as a loading wall. I looked at the specs for the 10w6v3...
