Replacing factory speakers?

I want to redo the whole sound system in my mustang. Problem is I'm not really sure what I need to know about replacing the factory speakers. Some of them are already blown and I wanted to replace them regardless.

Also, I'm trying to figure out what else I need and this is what I've got so far..

-250 watt rms amp (seems low to me idk..)

-JL Audio 10W3v3-4

W3v3 Series 10" 4-ohm subwoofer (would like 2 but thinking i only need 1).

Id seal the bolt hole up with Silicone and maybe look into placing some Poly Fill inside the Plastic enclosure and mount the new sub and run it...
