In a ported box, the box reaches maximum efficiency at the port's tuning, which is why the sub is still loud, even though it is barely moving near tuning freq. Placement isn't extremely critical... usually aim it towards the back of the car.

 
The output from the port is only the majority of the total output at and around tuning. The port will really only be adding a significant amount of output 1/2 an octave above tuning. Anything higher than that, and the port does not resonate as much which lowers your output from the port.

It is pretty critical in SPL setups but not too much in daily driver setups. Placement will almost always have an effect on frequency response but it's really hard to say how it will affect things without trying out all sorts of combinations.
