neg - (run) from front to rear

what kind of car?

If its Full frame, then grounding to the frame ion the rear should be ok, may not be optimal, but I doubt you would have any serious problems

If its unibody than Id def do a negative run front to back

 
how long should i go with a dry cell and wet cell batt connected and if i do a run from front to back should i also ground it to the chassis in the rear
you could groud it and the amp(s) to the same place, may help you out some
