If I were to change my door mounts without changing the factory head unit and not running the new door mounts on an amp, would there still be a major change in the sound or would I also need to run the door mounts on an amp as well to get a better sound?

by door mounts you must mean speakers presume. You will hear a difference in sound do to it being, most likely, a distinct tweeter and midrange. But running off the stock HU you might only be getting about 8 watts rms per speaker which will hold you back dramatically. Some stock HUs are more powerfull but I remember when I took my stock HU out of my old car it was only a 8watt per channel on it.
