well plywood is made from thin layers of wood layered in alternating directions and mdf is made from small pices of wood compressed together with glue and resins under very high pressures. mdf is better for building boxes though. so i guess thats the difference. oh ya plywood is actually stronger but i dont know why they always make boxes out of mdf. hope this helps

Plywood is harder and has the tendancy to create resonances when used in speaker enclosures. MDF is more acoustically neutral.
