Well, in order to have lift, you need air moving over and under the wings. The lift of an airplane is caused by the bernoulli principle. if you have a plane going at take off speed, riding on a treadmill that is going at take off speed in the opposite direction, you have no airflow over the wings, thus , you have no bernoulli principle, therefore no lift, therefore no flying.
