Subwoofer position

Does it madder where the subwoofer is positioned in an enclosure? If the vent style port is on the left side does it make a difference if the sub is in the center of the box or the far right side? Thanks!
It does. You want the sub as far away from the port mouth as possible, less turbulence.

 
Port mouth? Is this the port opening inside the box or outside? So if my rectangle box has the port on the far left side, the sub should be on the far right correct? Or if you mean inside port mouth then the speaker should be farther left on the box, since the port exits and on the left but starts on the right side inside the box. Am I getting this right?
