stinger sgp32 isolator

That is about as good as you will find. The largest I could find in the diode based units was around 200A, like this one. What I did learn is that you can parallel them just like batteries if you need more than 200A of throughput.
Just an FYI though, you will see a small drop in voltage using this style of isolator, so the MLA adapter or an external regulator would be helpful to offset the voltage loss.

Yes, the diode based units (look like heat sinks, not caps) will always keep them separate. They become two completely separate circuits.

The other style, the solenoid type that look like a small cap, they only isolate with the car off.
