caulk and silicon is the devil. i only us a little on my terminal cups. if your cuts a strights u wont need silicon.

ALRIGHT, all the boxes ive built sealed and ported are airtight. if you use enough glue shoulnt have to use silicon. a friend of mine uses a spray sealer, that actuly penatrates the MDF. its used for roofing.

 
I wouldn't call them a must...I also don't like just drilling a hole and sealing it. If you're not going to use a terminal cup, at least get a good set of binding posts and use those to attach your cables.
