Only if your amp is high current , and or stable at 2 ohms mono, 1 ohm stereo. If your unsure don't try it, the amp might shut down and possibly be damaged.

For example Phoenix gold, Soundstream, and JL Audio amps are high current. Kenwood, Fosgate, and Sony amps are not.

 
If the amp is rated 150x2 you can hook up one sub to each channel (stereo). Or you could bridge the amp (mono) to power one sub. I would just hook up one to each channel, It might not be as much power but it will sound cleaner and probably louder.

The question is confusing. Connect the 4" to the radio and use the rear amp channels for the sub. If the sub is DVC you can also use it in stereo...
