How difficult is it to make box in spare tire location.....

A Enclosure in the spare tire area of a 2006 Dodge Charger? I have a Kicker Comp Vr12 that I would very much like to put in that section of the trunk to open my trunk back up with space if needed to use it. I'm not all that into taking up the entire trunk space with a box and not being able to ever use my trunk ever again for groceries or what ever needed to store. Is this best left to the professionals to get it done right?

It'll be expensive and you'd better enjoy the smell/itching from working with fiberglass. Not difficult though. Layer up 1/2" or so of fiberglass in the spare tire well, cut and mount a baffle to that. Simple sealed box made out of the spare tire well. Get creative from there, but it's been done plenty, I've done it, and besides the cost and hassle of fiberglass it's a good option.
