Help with some building basics

Hi

I have not built a box yet and need some advice. I have been reading up for ages and even got some power tools a few years ago to build a box. All I need to get is a workbench.

I wanted to know a few things like:

1. Do I need to use clamps to hold the box together during the drying process or will the screws be sufficient to hold the box together and make a good strong bond?

2. How long does the wood glue take to dry?

3. Can I use regular silicon caulk(got plently lying around which I used in the bathroom and to seal an older box)? I have read somewhere that the gas/emissions could damage the sub.

I will probably ask a few more questions later but that's what I remember for now.
