If I run the front RCA to an amp for my front 3 ways can I use the Headunits front speaker wires and run tweeters of the headunit while using the front RCA at the same time?

So long as you have the tweeters appropriately crossed over somehow and don't try and run them full-range off of the deck then yes.

But why, if you've got a 3-way front stage, are you wanting to add another tweeter?

I like to balance out my extra bass with extra highs personal preference. Once I hook my speakers up to my amp i may decide not to do this.
