Your amp needs to be ~ 13V min. if car is on.
Watch it to see if there is voltage drop on the high notes.

Low voltage can cause problems
And I just stick the leads to the power and ground of the amp to test this?

Would I possibly need to check the current too?

And how do I check the resistance of the subwoofer?

Most amps if they drop below 12 volts go into protect.
