Single 12" ported. Sub position relative to port.

A friend and I will be building a custom box for my dv woofer 1.9 cubes after displacement at 32hz. The sub n port will be loading off the rear of the trunk for audible loudness preference. Does it matter to have the sub and port on opposite sides of the face of the box or could the sub be centered with the port next to it without a loss of quality n sound?

It won't make a difference if the sub is centered or on the far opposite corner.
