How to Calculate Tuning Freq. on a Center Port Box?

same method, mean free path down the center of the port. tuning is based on port area and length.

if you have two subs with a center port, keep the divider between the two ports so tuning is consistent. otherwise, you'll get interaction between the two subs.

 
As keep_hope_alive put it, always try to keep all drivers equal distance from the port opening, to keep tuning as even between the drivers as possible. Center ported boxes are actually much easier to keep uniform/consistently tuned than are side ported boxes.

Just remember that you'll have double the center width as you do the back half. Example: 4" wide center would then decrease to 2" on the rear turns. You'll also need a centered 45° "triangle" at the back wall do direct airflow accordingly.
