Can this be done with HU?

is it possible to run speakers off of normal wiring harness on a HU and run all 6 preamp outs at the same time? or does one cancel the other out?

Either or, but not recommended using your method.It may be possible through high level speaker input towards amplifier, yet, not recommended.
