question about voltage from stock HU output

I'm trying to add a bigger subwoofer to my car, my stock HU has a separate amp. I looked at the diagram for the amp and it has some input wires labeled like this

R(+) R(-) L(+) L(-), please take a look at the diagram, I am not sure if those are the audio signal wires going into the stock amp from the stock unit? I'm trying to get audio signal so that I can input them in to my 500/1 amp for my aftermarket sub. If I want to make sure they are the audio signal and measure them using a volt meter. Would the AC voltage increase if I increase the stock HU volume? thanks in advance for your help.
