question about setting gains

OK, so someone is trying to tell me to keep the subs hooked up when setting with a multimeter to get the correct amount of power. It has been stickied all over the net on different forums that the speaker was to stay disconnected. Can someone explain how the amp knows to set it at a certain load if no load is present (no speaker hooked up)?
