You measure voltage/resistance in parallel with the circuit of interest, and current in series...

put the DMM in directed voltage the one with ---, and put the terminals on the amp + and -, this is for control gain, you should look for some manuals in the amp section for do this right.

Car voltage is DC, amp output is AC, speaker impedance is tested in Ohm (should see the greek letter some where on the rotary switch)
