Are these supports really nessesary

Can I use any wood really? If its not circle like that will it effect my tuning at all? Probably a stupid question just making sure though.
It shouldn't make any noticeable difference to the tuning of the enclosure. They suggest round bracing because it helps the flow of the waves, flat edged supports make sound waves bounce back where as a round support the waves can flow around and past it to the port opening.

 
For that enclosure, nah, ya don't need any bracing for it.

If the parts are cut clean, true and square and assembled properly, I highly doubt you got anything to worry about. Cut it, build it, don't sweat it.

Its already cut. Cuts are very clean and line up almost perfectly

The bond is stronger than the wood itself and I highly doubt it exploded due to the pressure the subwoofer created. A good, well built enclosure with good bond joints, you need a 2lb hammer to beat it apart or drop it from two stories up.

Then it simply wasn't built properly.
To the threadstarter, no, those dowels will do next to nothing for structural support.

85% fact. I only say that because MDF is special in that it doesn't exactly have wood fibers (I'm talking about running the length of the board, not the composition of the wood itself), but it still holds true. Particle board is very hit or miss though. When I tested all the Titebonds, and Elmer's carpenter and probond, all 5 never broke at the joint when properly clamped.
