More cone are allows for more SPL... so yes it does matter.

Generally larger subs get louder and require more power all things being equal. They also require bigger boxes etc.

i'm planning on getting 2 Pioneer 12's TS-300R model and they are goin in my car which is a 2001 ford taurus SE and the subs handle 150 rms each, how much power should i give them each? will they be louder than my single 12'' RF punch stage 1 sub?

2 12's is a start in the right direction. I do not know your specs on the subs and all that good stuff but my guess would be yes it will be louder. But that does not mean that 2 12's will always be louder than 1.
