If I had a box that was 30X13.5X16 (outside) and was built with 3/4 MDF, would the internal volume be 3.3 cubic ft.? This is before sub and port displacement.

You have to double the thickness of the MDF since it exists on both sides of a dimension. 30" outside means 28.5" inner.

Yep...subtract 1.5 from every dimension and then go normally from there...
