50hz test tone

If I have 2 Fi Q 12" Subs in a 4.5 Cu.ft box tuned to 32hz would I use a 50hz test tone to set my gain? or do I find a tone thats close to my subs fs which is 28.4 per sub? I've searched and can't find a straight answer.
Since your setting your gains on the amp it really has nothing to do with that size enclosure you have, or what it's tuned to.

Thanks and just for my personal preference how come its always 50 or 60hz is it because thats usually where people set there LPF?
50 to 60 Hz cycles is the frequency range where most digital multi-meters are best at measuring AC current.

ETA: I forget the exact frequency that household AC current flows at but I think it is close to this range.
