Why when I turn my car on it make a loud humming noise out of my subwoofer?

There are several things that could cause that. First, disconnect RCAs at the amp and test it. Reconnect at amp, and disconnect at radio and test it. Tell us the outcome.

Yes, make sure your grounding point is clean and tight. If it still hums, connect your battery in the trunk direct to the amp, and test without RCAs. If it still hums after that, sweep every pot and press every switch a few times while the amp is on, but without RCAs.
