Im looking to build a box for my e-15 powered by a saz-1000d. I'm thinking 4 cubic ft tuned to 32hz. Im thinking of doing an aero port box. Would a six inch aero port being 15.21 inches long be the correct measurement?

Yes that is right, just make sure that your GROSS box volume takes the port and driver displacement into account....shound end up with ~4.38 cubic feet

This is going to be powered by a saz-1000d in a 4dr blazer. Where is the best place to put the port? On the passenger side of the box? Facing the rear?
