Tuning Questions >?????? Plus Polyfil question !

I am a little confused about tuning , Whats is the effect of tuning a box to 20 versus say 40 , What are the diff in sound coming out ?? What would a box do at 20 that it wont do at 40 ?? Is it that at 20 it just is louder on lower notes ?? or what , Please explain as best you can I have always ran sealed boxes and donth ave much experience with vented

The lowest of the low songs go into the low-30s/high-20s.
Ok how about this one , Should you use any polyfill in a vented enclosure ? If so how much ??
