Silicone is for sealing.
Wood glue is for bonding pieces of wood together.

I glued mine, while assembling it. Then sealed the inside seams with silicone.... Don't know if that is overkill or what....:D

Both.
Glue the joints, then seal the edges.

Either works just fine. When i built my first box, i used silicone. But, on my last one, i didn't have any silicone around, and was lazy, so i didn't feel like going to the store. so, i just used wood glue and sawdust from cutting the panels, and made a paste out of that. same way you can patch mdf and some other plys. worked just fine.

I really dont think it matters as long as it seals.
I use the **** from walmart for .99 cents to seal up any leaks i might have.
