Alright.. I'm doing a 25"x6" Aero... (other size suggestions are welcome) for a 2.5cube box @ 32hz..
Soo.. How will I brace the port? I think it will need more support than just at the hole it goes through..

Duct tape and guy wires?
Or you could box off a square section around the port made of MDF to support it.
this box just wont work 2.5 cubs and tuning that low just doesnt add up you would need 55" straight lenght to tune that low. i Suggest building a bigger box. What type of car and subs you using?

I was like huh my 2 cube at 43 is a 4" port
Forget what i said before i thought you were going to run 2 ports instead of 1. i still suggest a bigger box

That's the correct length. 25 1/4" should be almost dead on 32 hz.

According to the psp-inc calculator. 2.5 @ 32 hz and one 6" aero, needs 26.38" of length. So yea you are really close. 25.25" will get you a tuning of around 32.6, according to their calc.
