I got all the dimesions and port lenght for my box. The thing is I don't know what to use. I know you use 3/4 mdf but what type of glue, screws, and silicone sealent do you use?

Use liqiud nails on all the joints before you srew them together. For screws just use 1 5/8 wood screws and the ones you put in near the corners you may want to pre-drill.

Also if i am building a 4ft^3 box divided into 2 seperate chamber do I need to brace it internally? Dimesions are 35x14x24 I believe.

If you are separating it into two cambers you probably don't
need any additional bracing. It never hurts to brace though.

Even just a small piece of MDF from front to back can really help
reduce flex.
